ABSTRACT

In the present work we analysed the influence of orchiectomy and testosterone injection on soluble (S) and membrane bound (MB) aspartyl and glutamyl aminopeptidase activities (AspAP and GluAP, Aminopeptidase A) in serum, frontal cortex (FC), Hypothalamus (Ht), hypophysis (Hp) and adrenal glands (AG). Forty male mice (balb-c) were distributed in five groups: Control (C), orchiectomized (ORX), and orchiectomized and injected with different doses of testosterone (D1, D2, D3). In serum, an increase on AspAP and GluAP activities were observed in D1 and D2 groups. Soluble AspAP decreased in FC after orchiectomy. These lower levels were maintained after the injection of testosterone. In the other tissues analysed no changes were observed. Soluble GluAP activity decreased with orchiectomy in FC and Ht. A significant increase were observed in this activity in Hp of ORX animals. The administration of testosterone produced a decrease in Ht. In Hp and AG no changes were detected. No changes were observed after orchiectomy in membrane bound AspAP. In ORX animals a significant increase of MB GluAP activity were detected. These results indicate the existence of an hormonal influence on aminopeptidase activity. Due to the role of aminopeptidase A on the systemic and local renin-angiotensin systems, hormonal modifications during physiological and/or pathological conditions could modify blood pressure regulation through these systems.
